NAD+

NAD+ is the oxidized form of nicotinamide adenine dinucleotide, a dinucleotide of nicotinamide riboside and adenosine joined by a pyrophosphate bridge[1, 2]. It occurs in all living cells.

Structure and names

Modifications:

  • nicotinamide riboside and adenosine joined by a 5′-5′ pyrophosphate bridge
  • positively charged pyridinium nitrogen balanced by one phosphate negative charge (inner salt)

Stereochemistry: beta-D-ribofuranosyl units, (2R,3S,4R,5R) at both riboses

CAS note: 53-84-9 is oxidized NAD+ as the inner salt (zwitterion), sold as ‘free acid’. Sodium salt is 20111-18-6. Reduced NADH is 58-68-4 (C21H29N7O14P2, 665.4) and NADH disodium salt is 606-68-8 (709.4). Commercial lots may be hydrates.

Other names: nadide (INN, USAN), nicotinamide adenine dinucleotide, beta-NAD, NAD, diphosphopyridine nucleotide (DPN), coenzyme I, beta-NAD sodium salt (salt form)

PubChem CID 5892 · UNII 0U46U6E8UK[1, 3, 4]

Structural features:

  • Two nucleotides, nicotinamide riboside 5′-phosphate and AMP, linked through a pyrophosphate.
  • Oxidized pyridinium ring; reduction to NADH adds a hydride at the nicotinamide C4 position.
  • Net charge near neutral as the inner salt; often supplied as a sodium salt.
  • Highly polar and non-volatile; the pyrophosphate groups and adenine ring are ionizable, which affects retention.
  • The registered inner salt is drawn with one positive and one negative charge; at the pH of an analytical method the phosphate groups are further ionized, so the observed charge depends on conditions.

Research background

FDA lists NAD and NADH disodium in 503A bulk substance Category 1 (under evaluation) as of May 14, 2026[5].

  • NAD+ is a coenzyme for redox reactions, accepting a hydride to form NADH, and is central to energy metabolism. (review)[2]
  • NAD+ is consumed as a substrate by non-redox enzymes including sirtuins, CD38 and poly(ADP-ribose) polymerases. (review)[2]
  • Tissue and cellular NAD+ levels were reported to decline with age in rodents and humans. (review)[2, 6]
  • Precursors such as nicotinamide riboside and nicotinamide mononucleotide were studied in animals as ways to raise NAD+ levels. (review)[6]

Analytical considerations

  • NAD+ and NADH can interconvert during extraction, biasing measured NAD+/NADH ratios.[7]
  • Acid-catalyzed degradation occurs after acidic extraction; samples need prompt neutralization.[7]
  • Free acid, sodium salt and hydrate forms differ in MW; counterion and water content shift assay results on an as-is basis.[1, 3, 5]
  • NAD+-consuming enzymes cleave it to nicotinamide and ADP-ribose[2]; a method should retain and resolve these polar products.

USP–NF status

Sample and formulation

  • Store powder cold, dry and protected from light; NAD+ solutions are less stable than the dry solid.
  • Confirm whether the product is NAD+ or NADH, and free acid or sodium salt, before testing.
  • Supplied as lyophilized vials, bulk powder, capsules and sublingual or liquid products.
