Vitamin B12 (methylcobalamin)

Methylcobalamin is one of the two coenzyme forms of vitamin B12, a cobalt-centered corrin with a methyl group bound directly to cobalt. Vitamin B12 is a family of cobalamins; methylcobalamin is distinct from cyanocobalamin, the cyanide-bound form.

Vitamin B12 (methylcobalamin) testing at Vanguard Laboratory: purity, quantity and identity from $250 per sample. Results in 3 to 5 business days; sterility 14 days.

Full USP monograph testing is available (USP monograph: Methylcobalamin). See USP Testing.

Structural features:

  • Corrin macrocycle with a central cobalt(III) ion
  • Cobalt-carbon (Co-CH3) bond, which is photolabile
  • Lower axial 5,6-dimethylbenzimidazole nucleotide with a phosphodiester link
  • Large cobalt complex (about 1344 Da) that is red in solution

Research background

Sold in the US as a dietary supplement ingredient covered by USP dietary supplement monographs[2, 4]; listed as mecobalamin in the Japanese Pharmacopoeia (JP19)[3].

  • Methylcobalamin is the cofactor for methionine synthase, which remethylates homocysteine to methionine using 5-methyltetrahydrofolate as the methyl donor. (review)[5]
  • Adenosylcobalamin, the other coenzyme form, serves methylmalonyl-CoA mutase in mitochondria; the two forms have distinct enzyme roles. (review)[5]
  • Vitamin B12 deficiency was reviewed as causing hematologic and neurologic disorders linked to these two enzyme pathways. (review)[6]

Summaries of published research, for reference only. Not a claim of safety or efficacy.

Analytical considerations

  • The Co-methyl bond is photolabile; in aqueous solution at pH 7.4 under experimental UVA exposure, methylcobalamin converted to hydroxocobalamin within seconds, so samples need light protection.[7]
  • The assay must separate methylcobalamin from hydroxocobalamin and cyanocobalamin; USP lists both as reference standards for the methylcobalamin monographs, which establishes reference material, not a given laboratory’s method.[2, 4]

Sample and formulation

  • Protect from light at every step: amber containers, foil wrap and subdued lighting during preparation.
  • The USP assay is calculated on the anhydrous basis, so water content affects reported purity.
  • Common formats: tablets, sublingual lozenges, capsules, liquids and injection vials.
